Progress in the Development of Lactococcus lactis as a Mucosal Vaccine Delivery Vehicle
نویسندگان
چکیده
Lactococcus lactis, a food-grade, non-pathogenic, non-invasive, non-colonizing and “generally regarded as safe” lactic acid bacteria, is widely used in food, medicine, and husbandry industry, and it is a potential and promising candidate as a mucosal vaccine delivery vehicle (MVDV). This review describes the latest research progress of L. lactis as an MVDV and its potential improvements. Firstly, the review introduces the advantages of using L. lactis as an MVDV, emphasizing the efficient controlled protein expression and protein-targeting systems developed for production of a desired antigen. Secondly, an extensive overview is given of the progress made in improving production yield and stability of the heterologous proteins expressed in L. lactis. Thirdly, an overview is provided of the efficiency of L. lactis as MVDV for mucosal immunization. Finally, the problems limiting the use of L. lactis as MVDV are introduced and probable methods to solve them are brought forward. _____________________________________________________________________________________________________________

Lactococcus lactis as a live delivery vector
Mucosal surfaces of the body provide a universal entry portal for all known and emerging infectious pathogenic microbes. Therefore, it seems that special vaccination strategies are needed for vaccines that can hinder the entry capability of pathogenic microbes through the mucosal surfaces. Lactic acid bacteria are widely used in the food industry and are presently applied as delivery vehicles i...
متن کاملHeterologous protein production and delivery systems for Lactococcus lactis.
Lactic acid bacteria (LAB), widely used in the food industry, are present in the intestine of most animals, including humans. The potential use of these bacteria as live vehicles for the production and delivery of heterologous proteins of vaccinal, medical or technological interest has therefore been extensively investigated. Lactococcus lactis, a LAB species, is a potential candidate for the p...
متن کاملInduction of Immunogenic Response in BALB/c Mice by Live and Killed Form of Recombinant Lactococcus lactis Displaying EG95 of Echinococcus granulosus Immunogenic Response in BALB/c Mice
Background: CE is a zoonotic parasitic infection caused by Echinococcus granulosus worldwide and is associated with economic losses among livestock animals. EG95 is an immunogenic antigen from the E. granulosus. Lactococcus lactis has been prested as a safe vehicle for antigen delivery. The goal of this study was to design a novel L. lactis strain displaying EG95 as a vaccine delivery system. M...

Bacterial-based systems as live vectors for the delivery of heterologous antigens offer a number of advantages as vaccination strategies. Developments in genetic engineering have given Gram-positive lacticacid bacteria (LAB) the advantage of being used as a host expression system for antigen delivery to inducethe immune response. A fragment containing the full length of the “eprA1” ...
متن کاملBroadly protective immunity against divergent influenza viruses by oral co-administration of Lactococcus lactis expressing nucleoprotein adjuvanted with cholera toxin B subunit in mice
BACKGROUND Current influenza vaccines need to be annually reformulated to well match the predicated circulating strains. Thus, it is critical for developing a novel universal influenza vaccine that would be able to confer cross-protection against constantly emerging divergent influenza virus strains. Influenza virus A is a genus of the Orthomyxoviridae family of viruses. Influenza virus nucleop...
